Microscope Slide Abbreviations
         Stain and Observe Cheek Cells
Cells from the lining of the mouth provide an easy source of eukaryotic cells for observation and are an economical choice for fun in the laboratory . Plus, cheek cells are a great way to connect textbook drawings and models to real life! Follow this simple procedure:
1 . Use a fresh, sterile cotton swab for each preparation . Immediately after use, place the cotton swab into a biohaz- ard bag or a beaker of bleach solution . Do not place items on the lab table .
2 . Wash hands thoroughly with soap and water before begin- ning the lab activity .
3 . Use lens paper to clean a microscope slide .
4 . Using a sterile cotton swab, gently rub the inside of the cheek . Be careful not to irritate the epithelial tissue or to draw blood .
5 . Rub the collected material onto the microscope slide in an area about the size of a dime .
6 . Allow the slide to air dry for one to two minutes .
7 . Carefully place one drop of the stain onto the sample area .
8 . Carefully lower the coverslip at an angle over the wet sample
to minimize air bubbles .
9 . Observe the slide preparation . If the stain is too dark for easy observation, carefully add a drop of water along one edge of the cover slip . To draw the water through the slide, place the edge of a piece of paper towel on the opposite edge of the cover slip .
10 . After completing the observations, place the microscope slide into a biohazard bag or a beaker of bleach solution .
