In this experiment, restriction enzymes are introduced as a tool to digest DNA at specific nucleotide sequences. Bacteriophage lambda DNA has a linear structure and multiple recognition sites. Separation by agarose gel electrophoresis of digests of lambda DNA will yield separate bands corresponding to the DNA fragments. Students estimate the size of the fragments and determine which restriction enzyme was used to digest the DNA. Kit includes detailed instructions and enough materials for eight gels. Electrophoresis apparatus, power supply, micropipets and tips, balance, microwave or hot plate and a white light illuminator are required and available separately.
Note: DNA is included as Ready-to-Load QuickStrip™ connected tubes or individual 0.5- or 1.5-mL microcentrifuge tubes. While these samples are room-temperature stable for up to one month after receipt, a refrigerator is recommended for long-term storage.

Materials Included in Kit (for 8 gels): Instructions, InstaStain® Blue Cards, FlashBlue™ DNA Stain, Ready-to-Load DNA samples, calibrated pipet, UltraSpec–Agarose™, electrophoresis buffer (50X), disposable pipets, practice gel loading solution.
Additional Materials: DNA electrophoresis apparatus, microwave or hot plate, white light illuminator, 5–50 µL micropipets (optional).
