Hide and Seek: A Coliform Test—

The unique indicator dyes in the media make counting coliform colonies easy . The open-ended inquiry activity in this kit has students test items of their own design for coliform presence . Your students will be amazed to find out that water that looks clear may be harboring dangerous pathogens . Students thoroughly investigate and compare several antiseptic and disinfectant products, such as soap, alcohol and bleach . With the buzz in the media about bacterial resistance, you will definitely hold a captive audience while discussing this important topic! Student instructions, worksheets and Instructor’s Notes included .
Complete for 30 students working in pairs. Bacillus megaterium (LM1002) is required and available separately.
Transformation of E. coli with pGalTM (Blue Colony)TM Transformation involves the uptake of extra-cellular DNA by another cell, resulting in a newly acquired genetic trait . In this experiment, the bacterial “transformants” acquire antibiotic resistance and a blue color, following uptake of plasmid carried beta-galactosidase and ampicillin resistance genes . The plasmid used, pGalTM, is unique, and designed specifically to simplify the process . Cells provided are freeze-dried and ready to reconstitute and use—eliminating the need for costly overnight shipping and preparation of competent cells . Transformation and plat- ing of cells requires 50 minutes—followed by overnight incubation .
Kit contains materials sufficient for 10 lab groups—with each group preparing these cell platings.
