Kastle-Meyer Reagent—Forensic Test
Classic test for the presence of blood at a crime scene! Kastle-Meyer reagent contains reduced phenolphthalein indicator in a basic solution. Simply wipe a presumptive bloodstain with a cotton swab, add a drop of the reagent, and then apply a drop of hydrogen peroxide. If the swab turns pink rapidly, it is positive for blood! For a terrific guided-inquiry forensic activity, use the Kastle-Meyer reagent to investigate substances that will interfere with the test to give false positives.

Who’s the Daddy? DNA Profiling—

Discover the power of DNA profiling (fingerprinting) using this creative simulation. No expensive equipment or chemicals needed! This activity only requires paper and scissors. Students solve a paternity case as they re-create each step of the DNA fingerprinting process. After assembling double-stranded DNA “molecules,” students cut them into fragments to simulate restriction enzyme digestion and then separate the fragments by size in a simulated gel. Next the DNA fragments are “denatured” (cut into single strands), “blotted” (pattern copied onto a sheet of paper), and treated with simulated radioactive probes that combine with only specific base sequences. The resulting simulated autoradiograph iden- tifies target DNA fragments, which are used to match the unidentified parent. This is a great teaching tool for students to learn how DNA fingerprinting really works! Includes black-line masters of simulated DNA segments, “radioactive probe” locators, electrophoresis gel blot- ter, magnetic tape, and zipper-lock bags.

An introductory activity for students about to begin the study of biotechnology and electrophoresis. Students become medical forensic investigators as they read a case in which a woman has been attacked or a case in which an elderly man has been murdered. Once the students have read one of the case reports, they are provided with photo- reproductions of actual electrophoresis gels. Students will interpret the evidence against a list of possible suspects and attempt to solve the mystery. The design of the activity affords the teacher the flexibility to alter case outcomes from class to class and even manipulate evidence. Kit includes in-depth background material, lab preparation and instruc- tion tips, student copies of case descriptions and investigation reports, and reproductions of evidence and suspect electrophoresis gels. The curiosity and excitement this activity creates will lead to a better under- standing of electrophoresis and biotechnology. Time required for this activity is 30 minutes.
