A fun kinesthetic way to teach DNA structure and restriction enzymes. Every student in your class becomes a coded nucleotide, and together the entire class will form a giant DNA molecule. Students will person- ally experience the antiparallel and coding structure of DNA. Once built, the giant DNA molecule is cut by a restriction enzyme. Then the challenge for each DNA fragment team is to run an obstacle course simulating gel electrophoresis. This DNA activity will be remembered! A Flinn workshop favorite! Includes base pair connecting rods and complete teaching instructions.

Gel Electrophoresis Simulation—
Super Value Kit

This pop bead simulation will help students visualize how the sepa- ration of molecular fragments occurs during gel electrophoresis. This involving set of materials is a great precursor to actual electro- phoresis work. Students build models of DNA fragments with pop beads and then place them on a paper simulated agarose gel. Based upon the fragment length and migration distance, students calcu- late the Rf values and draw a standard curve plot for their sample. From the standard curve plot they can deduce the length of an unknown fragment. Includes over 600 pop beads. The provided masters allow students to complete the standard curve plots individually. Six different sets of fragment data are provided so that student groups can be working on different samples during the lab.

A reality-based exploration into identity testing using mitochondrial DNA analysis. Your students play the role of investigators in an abduc- tion case. The fascinating and highly educational simulation combines elements of history, human rights, family impact, forensic science, and genetics.
In the course of their investigation, students will:
• “Digest” short, 60 base-pair sequences of mitochondrial DNA using
Hae III restriction enzyme.
• Determine the sizes of the resulting “fragments” and use a standard
curve to plot the migration distance for each fragment on a paper
“gel”.
• Label the separated fragments with a “radioactive probe sequence”.
• Interpret the results to determine the true parentage of a child alleg-
edly abducted as an infant.
Includes extensive background information, objectives, reproducible student copy masters, detailed results, study and follow-up questions, glossary, and extension ideas. The activity is designed to be conducted by cooperative teams and can be completed in two class periods.
