Electrophoresis and DNA Forensics— Laboratory Kit

The world of forensic science was revolutionized with the discovery of scientific techniques for identifying humans, and all living things, using DNA. DNA fingerprinting can be used to identify the source of DNA in forensic investigations and has also been used to diagnose genetic diseases, identify disaster victims and study evolutionary relationships among organisms. In this activity, students investigate the basic prin- ciples of DNA forensics using gel electrophoresis, which allows them to identify matching DNA profiles from a collection of DNA samples.*
Complete for 24 students working in pairs. Electrophoresis units and power supplies are required and available separately.
Morphology of Cancer Cells
Your students become pathologists when they fix, stain, and mount specimens of both cancer cells and normal cells. Cancer cells show many unique features when compared to normal cells. Some of these differences are readily visible under a light microscope with a 40X objective! Cell morphology and cell density changes are easy to see as each multisport slide contains separate areas of each cell type. Complete for 6 student groups. The entire laboratory activity may be completed in one class period. Microscopes with 40X magnification are required and available separately.
Blue/White Cloning of a DNA Fragment—
Student Laboratory Kit
Blue/white screening is one of the ways scientists determine if cloning has been successful. In this kit, students complete a ligation, a transforma- tion, and an assay, which is based upon the blue/white colony screening protocol. If the gene that expresses ß-galactosidase is interrupted by the cloning plasmid, then white colonies will grow because the enzyme is not able to cleave the X-Gal indicator. If the ß-galactosidase gene is not cut by the plasmid, then the X-Gal is cleaved into galactose and a blue indigo compound. So, if the colonies are blue, then the insertion failed. Three convenient halt points are indicated in the procedure so the experi- ment can be performed over several days in three 60–70 minute sections.
Complete for 5 groups of students. Includes DNA fragment, linear plasmid, T4 DNA Ligase, competent cells X-gal, and all necessary reagents. An incubator, two waterbaths, a shaking incubator or shak- ing waterbath, microwave or hot plate, automatic micropipet and tips, spectrophotometer, balance, centrifuge, and microcentrifuge are all required and available separately.*‡
C. elegans Ecology Experiment Kit
In as little as two weeks your students can become working scientists using the model organism Caenorhabditis elegans. Because this small nematode—1-mm long as an adult—responds like a higher order animal, it is used in many industries and research facilities as the test subject for toxicity, genetic, and behavioral studies. In this experi- ment, students use pre-diluted heavy metal solutions to determine the behavioral effect of heavy metals on a normal C. elegans versus that of a toxicity mutant C. elegans. After becoming experienced culturing this model organism, your students may come up with some interesting experiments of their own. A great opportunity for inquiry!
Includes all heavy metal solutions and necessary lab supplies. A redemp- tion card is included for all perishable material. Please allow 2–3 weeks for delivery of perishable materials after sending in the redemption card. Microscopes and a water bath are required and are available separately.
