In this lab experience, students carry out an investigation to determine the sex of fruit flies as well as common mutations. Students use drawings of Drosophila melanogaster as well as live specimens to get hands on experience with genetic terms, mutations and monohybrid crossings. Using live fruit flies, students learn the technique to anesthetize the flies as well as separate them by gender and eye color mutation. Students conduct a monohybrid cross to observe the recessive inheritance pattern of the sepia body color in the F1 generation and make a prediction for the F2 generation. Wild-type and sepia Drosophila are required and available separately.
Editable, differentiated instructions range from a time-sensitive prescriptive lab to full open inquiry, and robust online videos and content—including a virtual reality (VR) simulation—help students prepare for and better understand the labs they’re conducting.

Concepts: Genetics, monohybrid cross, genotype vs. phenotype
Outcomes: Students are able to easily identify the differences between male and female flies as well as identify which ones are homozygous recessive for sepia and which are wild type fruit flies. They will gain an understanding of why scientists use fruit flies for genetic studies and how mutated traits are passed from generation to generation.
