In this lab experience, students will experiment with temperature variances as an environmental factor that impacts gene expression in the bacteria Serratia marcescens. Students will also have the opportunity to test the antifungal properties of Serratia marcescens against a common fungus associated with food spoilage, Rhizopus stolonifer. Serratia marcescens is required for all lab levels and available separately. Rhizopus stolonifer is required for the advanced lab and available separately. Editable, differentiated instructions range from a time-sensitive prescriptive lab to full open inquiry, and robust online videos and content—including a virtual reality (VR) simulation—help students prepare for and better understand the labs they’re conducting.

Concepts:
Microbiology, gene expression, transcription, translation, enzymes 
Outcomes: Students will discover that the Serratia marcescens bacteria grown at room temperature or about 25 °C will result in smooth, dark red bacterial colonies. The bacteria at 5 °C are grown at a temperature in which the genes are expressed, and the enzymes responsible for the synthesis of prodigiosin are produced. The cultures grown at 37 °C will display white or pale pink colonies.
