Students solve a paternity case as they re-create each step of the DNA fingerprinting
process. After assembling double-stranded DNA “molecules,” students cut them into
fragments to simulate restriction enzyme digestion and then separate the fragments
by size in a simulated gel. Next the DNA fragments are “denatured” (cut into single
strands), “blotted” (pattern copied onto a sheet of paper) and treated with simulated
radioactive probes that combine with only specific base sequences. The resulting
simulated autoradiograph identifies target DNA fragments, which are used to match
the unidentified parent. Includes black-line masters of simulated DNA segments,
“radioactive probe” locators, electrophoresis gel blotter, magnetic tape and zipperlock
bags.

with photo-reproductions
of actual electrophoresis gels. Students will interpret the evidence against
a list of possible suspects
and attempt to solve the mystery. The design of the activity
affords the teacher the flexibility to alter case outcomes from class to class and even
manipulate evidence. Kit includes in-depth background material, teacher preparation
and instruction tips, student copies of case descriptions and investigation reports,
and reproductions of evidence and suspect electrophoresis
gels.

Every student in your class becomes a coded nucleotide, and together the entire
class will form a giant DNA molecule. Students will personally experience the
antiparallel and coding structure of DNA. Once built, the giant DNA molecule is cut
by a restriction enzyme. Then the challenge for each DNA fragment team is to run
an obstacle course simulating gel electrophoresis. Includes base pair connecting
rods and complete teaching instructions.

Students build models of DNA fragments with pop beads and then place them
on a paper simulated agarose gel. Based upon the fragment length and migration
distance, students calculate the Rf values and draw a standard curve plot
for their sample. From the standard curve plot they can deduce the length of an
unknown fragment. Includes over 600 pop beads. The provided masters allow
students to complete the standard curve plots individually. Six different sets of
fragment data are provided so student groups can be working on different samples
during the lab.
