Identifying Proteins and

In this exploratory student activity, students first compare and contrast the behavior of
three common proteins—albumin, casein and gelatin—using the biuret test to identify
the general nature of the peptide linkage and its central role in protein structure.
Students then perform a series of chemical tests to further explore the composition
of these proteins with respect to the presence of three specific amino acids—arginine,
cysteine and tyrosine. By studying the distribution of different amino acids in proteins,
students learn how hydrophobic and hydrophilic amino acid side chains stabilize
proteins and contribute to their special properties. Includes reproducible student
handouts, background information, Teacher Notes with sample data and answers to
all questions and all necessary chemicals and consumable supplies needed.

In this hands-on kit, students examine the effects of acids and bases, inorganic salts,
organic solvents and temperature on the physical properties of proteins. Students
learn how these factors affect the interactions among amino acid side chains that
are responsible for the stable three-dimensional structures of proteins—and thus
change the actual shapes of proteins. Students observe that some changes are
easily reversed and thus relatively harmless while other changes are essentially irreversible
and lead to complete loss of protein function. The definition and applications
of protein denaturation are also explored. Includes reproducible student handouts,

use Sudan III—a special “fat stain”—to identify lipids. Students then perform a test
for unsaturation on a variety of seed oils to learn more about the vital role of saturated,
unsaturated and polyunsaturated triglycerides in nutrition and health. Finally,
students extract peanut oil from peanuts using hexane to determine the amount
