First students simultaneously run two identical DNA samples, one cut with EcoRI
and the other cut with HindIII, using gel electrophoresis. Then the banding patterns
produced are analyzed to see how the use of different restriction enzymes results
in changes in the number and size of the bands. This lab helps students understand
the theory of how restriction enzymes digest DNA. It also enhances laboratory skills,
such as preparing an agarose gel, loading and running DNA samples and staining
and destaining the gel to observe the separated bands of DNA.
Materials provided include sufficient agarose to make 6 gels, DNA samples cut
with EcoRI and HindIII, TAE buffer and methylene blue for staining the gel as well as
needle-tip pipets and large weighing dishes. Complete for 6 student groups. DNA
source is Lambda phage.*

A disease known as familial hypercholesterolemia (FH) causes an increase in blood
levels of the “bad” form of cholesterol, known as low-density lipoprotein (LDL). In
heterozygous patients, untreated FH causes a shortened life span, while homozygous
individuals usually die before age 20 of coronary heart disease. In this experiment, a
simulated genetic test for FH is demonstrated in which patients are tested for a DNA
polymorphism linked to the FH gene.
The experiment requires electrophoresis apparatus, power supply, balance and
a transilluminator. The kit contains complete instructions, background information,
ready-to-load DNA samples, agarose, buffers and the necessary stains. DNA source
is Lambda phage.*

Improperly loading will adversely affect the experiment. In this kit, the wells are
constructed of a unique gel material that is not easily destroyed by micropipet
tips. The tough polymer material can also be rinsed and reused many times. The
well holes are slightly oversized so students will be successful with their first pipet
transferring experience. The reusable quality makes them a real bargain!

electrophoresis apparatus. The color changes at the two ends of the electrophoresis
tank will be very striking when no buffer is used. When you add buffer solution

why a buffer solution is used in an electrophoresis apparatus. Enough materials are
