In the course of their investigation, students:
• “Digest” short, 60 base-pair sequences of mitochondrial DNA using Hae III restriction
enzyme.
• Determine the sizes of the resulting “fragments” and use a standard curve to plot
the migration distance for each fragment on a paper “gel.”
• Label the separated fragments with a “radioactive probe sequence.”
• Interpret the results to determine the true parentage of a child allegedly abducted
as an infant.
Includes extensive background information, objectives, reproducible student copy
masters, detailed results, study and follow-up questions, glossary and extension
ideas. The activity is designed to be conducted by cooperative teams and can be
completed in two class periods.

Students will simulate an important part of the engineering process as they manipulate
simulated plasmids. Following specific restriction enzyme cuts in their plasmid
models, they consider all of the possible recombination patterns that can result from
their plasmid pieces. All of the combinations represent potential newly-engineered
plasmids with unique characteristics. The key concepts related to recombinant
DNA will be visible and manipulated by students during this activity. Teacher Notes,

Recombinant DNA—

Students will construct plasmid DNA using chenille wire and four different colored
pony beads—each representing a different nitrogenous base. Students will manipulate
the plasmid by locating restriction sites and cutting the chenille wire as the
restriction enzyme would do to the plasmid. They will then insert genes of interest
using pony beads, and twist the wire back together to simulate ligating the plasmid.
Includes a prelab, paper-and-pencil activity to familiarize students with the process
of forming recombinant DNA molecules before beginning the activity. Includes reproducible
student handouts as well as detailed Teacher Notes.
